Transferring the cultured anther to a medium without activated charcoal overcomes the recalcitrance in pepper genotypes

Anther culture is a relatively easy and high-efficiency technique; however, low efficiency in plant regeneration may restrict its use crop breeding. Activated charcoal often used vitro culture, it ameliorate or hinder growth depending on genotype tissue used. Pepper one of the main vegetable crops Solanaceae family, but some pepper genotypes are known to be recalcitrant androgenesis formation haploid regenerants. Therefore, this study was aimed explore effect activated response genotypes. The material included 34 Long Green (LG), 13 Bell (BP), Charleston (Ch), 6 California Wonder (CW), 23 Capia (CP) advanced breeding lines. Initially, anthers were cultured medium with (WAC) for 25, 35, 45 d, then they transferred same without (NAC). In WAC medium, 15 lines LG showed highest recalcitrance while many CW had lowest androgenesis; after transferring 35-d-old NAC observed results indicated that from ideally 35 overcame pepper.
